With respect to National & Regional universities, there is no fundamental difference, it's just US News decides to organize their rankings this way. Regional universities get most of their students from the surrounding area, because they aren't so great that people move to attend them. Of course, top regional universities are very much lower in people's estimation that the top national ones, but they can be very good if you care about the education received rather than the brand. Even medium-ranked national universities have a better brand. In general, the most prestigious US universities are considered “National” ones, but there are hundreds of great regional universities that may suit highly qualified students well and may be more affordable and/or closer to home (which can reduce things like travel costs).
